摘 要:探讨了3种不同的缓冲体系和6种常见的金属离子对漆酶催化活性的影响。结果表明:不同的缓冲体系对漆酶催化活性的影响较大。在pH为2.0的磷酸氢二钾-柠檬酸缓冲体系(0.1 mol/L)中漆酶的催化活性最大。金属离子对漆酶催化活性的影响与金属离子的种类和浓度有关,在浓度低于0.1 mmol/L时,Pb2+、Ag+、Hg2+对漆酶的催化活性起激活作用,高于0.1 mmol/L时起抑制作用。在浓度低于20 mmol/L时,Mg2+、Zn2+、Cu2+对漆酶的催化活性起激活作用,高于20 mmol/L时起抑制作用。The effects on ABTS, oxidation catalyzed by laccase with different buffer solutions and metal ions ( Mg^2 + , Zn^2+ ,Cu^2+ , Pb^2+ , Ag^+ , Hg^2 + ) were investigated by Spectrophotometer. The results show that the best suitable buffer is K2HPO4- C6H8O7 buffer solution and the optimal pH was 2.0. The effects of metal ions on laccase activity are related to the type and concentration of metal ions. Pb^2+, Ag^ + , Hg^2 + activated the activity of laccase when the concentrations are lower than 1 mm, however, once the concentrations were higher than 1 retool/L, the activity was inhibited. As for Mg^2+ , Zn^2+ , Cu^2 + , the inhibition was cocurrcd only when the concentrations were higher than 20 mmol/L.
