摘 要:[目的]为油菜潮霉素抗性基因的遗传转化提供依据。[方法]以油菜品种沪油19号、苏州青、五月慢的子叶和下胚轴为外植体,研究不同浓度潮霉素处理对其褐化率、出愈率的影响。[结果]潮霉素浓度为3 mg/L时,五月慢、沪油和苏州青的褐化率分别为87.6%、30.7%、40.1%;褐化的临界浓度均为6 mg/L。苏州青下胚轴对潮霉素的耐受性较强,其褐化的临界浓度高于沪油19号和五月慢。潮霉素浓度为6 mg/L时,沪油19号、苏州青、五月慢子叶的出愈率分别为3.7%、2.3%、0,下胚轴的出愈率分别为5.1%、8.1%、2.2%。[结论]以子叶为外植体时,3个品种适宜的潮霉素筛选浓度为6 mg/L;以下胚轴为外植体时,沪油19号和五月慢适宜的潮霉素筛选浓度为6 mg/L,苏州青为9 mg/L。[ Objective ] The aim was to provide the basis for genetic transformation of hygromycin resistance gene of Brassica napus L. [ Method] With cotyledon and hypocotyl of B. napus varieties (Huyou 19, Suzhouqing, Wuyueman) as explants, the effects of treatments with different concn, of hygromyein on its browning rate and callus rate were studied. [ Result] When hygromycin, was 3 mg/L, the browning rates of Wuyueman, Huyou 19 and Suzhouqing were 87.6% , 30.7% and 40. 1% resp. and the critical concentration, they browning was 6 mg/L. The tolerance of hypocotyl of Suzhouqing to hygromycin was stronger and the critical concentration of its browning was higher than that of Huyou 19 and Wuyueman. When hygromycin, was 6 mg/L, the callus rates of cotyledon of Huyou 19, Suzhouqing, Wuyueman were 3.7% , 2.3% and 0 resp. and those of hypocotyl were 5.1% , 8.1% and 2.2% resp. [ Conclusion] When cotyledon of B. napus was taken as explants, the optimum screening conch, of 3 varieties on hygromycin was 6 mg/L, when its hypocotyl was taken as explants, the optimum screening conch, of Huyou 19 and Wuyueman on hygromycin was 6 mg/L and that of Suzhouqing was 9 mg/L.
