摘  要:目的以市售依托泊苷注射液和普通依托泊苷脂质体为参比制剂,评价了隐形依托泊苷脂质体在家兔体内的药动学。方法家兔单剂量静脉注射依托泊苷1.5mg/kg,于不同时间点采血测定,并计算药动学参数。结果隐形依托泊苷脂质体、依托泊苷注射液和普通依托泊苷脂质体的分布半衰期(t1/2α)分别为(2.05±0.32)、(0.05±0.01)和(1.73±0.26)h,消除半衰期(t1/2β)分别为(19.26±3.16)、(0.94±0.21)和(7.99±1.36)h,药时曲线下面积(AUC)分别为(26.04±3.53)、(0.98±0.26)和(11.65±1.70)μg·h/mL。结论隐形依托泊苷脂质体显著延长了依托泊苷在血液中的循环时间,具有缓释作用。Objective The pharmacokinetics of stealthy eoposide Iiposomes in rabbits was evaluated by comparison with etoposide injection and conventional etoposide liposomes, respectively. Methods Following intravenous injection administration at a dose of etoposide 1.5 mg/kg, the three kinds of etoposide preparations, the blood concentrations were measured at different time point and the pharmacokinetics parameters were calculated. Results t1/2a were (2.05±0.32), (0.05±0.01), (1.73±0.26) h, and t1/2a were (19.26±3.16), (0.94±0.21), (7.99±1.36) h, and AUC were (26.04±3.53), (0.98± 0.26), (11.65±1.70) μg.h/mL, for stealthy eoposide liposomes, etoposide injection and conventional etoposide liposomes, respectively. Conclusion The stealthy etoposide liposomes could extend significantly the duration of etoposide in blood circulation and contained characteristic of sustained-release.
