摘 要:对中国主要养殖的尼罗、奥利亚、莫桑比克、杂交种尼奥(尼罗×奥利亚)、奥尼(奥利亚×尼罗)和红罗非鱼(莫桑比克×尼罗)的线粒体COI基因的部分序列进行了PCR扩增、克隆和测序分析,探讨罗非鱼种间亲缘关系和种类鉴别标记。6种罗非鱼中大部分个体均获得长586-708bp的基因序列。其碱基组成GC含量为47.7%。序列分析表明,莫桑比克罗非鱼2个个体分为2个单倍型,单倍型之间的碱基差异为1个碱基,其余几种罗非鱼均只有1个单倍型。其中,尼罗和尼奥、奥利亚和奥尼的单倍型序列相同,但种间核苷酸序列差异达4.3%-7.7%。根据聚类分析结果可将这6个(杂)种分为3个组:尼罗-尼奥,红罗-莫桑比克和奥利亚-奥尼罗非鱼组。其中前2组亲缘关系较近,与后1组亲缘关系较远。表明C01可作为罗非鱼种类判别和亲缘关系分析的重要标记。Mitochondrial COI gene sequence variation of Nile (Oreochromis niloticus),Blue (O. aureus), Mozambique (O. mossambicus), Ni-ao (O. niloticus ♀ X O. aureus ♂), Ao-ni (O. aureus ♀ XO. niloticus♂) and red tilapia (O. mossambicusXO, niloticus) was investigated in order to understand the genetic relationship among them as well as to provide interspecific molecular markers for breeding practice. As a result,the partial sequence of COI was 586-708 bp long. The base compositions with GC content was 47. 4%. Sequence analysis indicated that Mozambique tilapias had two haplotypes and the other five species each had one haplotype. Yet there was only one base substitution between haplotypes in Mozambique and no mutation was found between individuals within the other species. However, sequence divergence between species was high up to 4. 3%-7.7%. Phylogenetic analysis indicated that the six species can be divided into three groups, among which the group of Nile and Ni-ao tilapias was closely related with the group of Mozambique and the red tilapias,whereas group of blue and Ao-ni tilapias was genetically distinct from the former two groups. These observations showed that COl sequence marker can be applied to species discrimination and phylogenetic study on tilapias.
