摘  要:[目的]研究用ISSR标记分析甜荞栽培品种的遗传多样性。[方法]采用ISSR分子标记对来自11个省的90份甜荞种质的遗传多样性进行了分析。[结果]19条ISSR引物共扩增出508条条带,平均26.7条;其中多态性条带462条,平均24.3条,多态性带的百分率为90.1%。Nei’s遗传相似系数在0.67~0.95。UPGMA聚类分析显示,聚类结果与地理来源有较强的一致性,来自辽宁、内蒙古、陕西、四川、甘肃等省的甜荞都是按来源各自聚为一类,但是发现了几个特殊的类型,即来自河北的甜荞E、安徽的82F以及辽宁的辽荞37号均单独聚为一类。不同地区材料之间的多态性信息指数(PIC)差异较大,其中,辽宁的最高,为0.842,内蒙古的最低,为0.633。[结论]采用ISSR分析甜荞遗传多样性,甜荞表现出丰富的多态性,表明可以用ISSR标记对甜荞进行分子水平的鉴定和遗传多样性分析。[ Objective] The research aimed to study the genetic diversity in common buckwheat( Fagopyrum esculentum Moench) based on ISSR markers. [Method] The genetic diversity of 90 common buckwheat accessions was analyzod by using ISSR markers. [Result] With 19 ISSR primers, totally 508 bands were detected,among which 462 bands were polymorphic,and the average number of band per primer was 26.7. The Nei's similarity coefficient of 90 accessions ranged from 0.67 to 0.95. Cluster analysis based on ISSR data showed that the accessions could be categorized into 4 major groups and 3 individual genotypes. The "common buckwheat E" from Hebei, "82F" from Anhui and "Haoqiao37" from Liaoning showed great genetic difference from other materials. There were obvious genetic difference among the landraces from Hebei, Anhui and Liaoning, and high similarity among cuhivars collected from different provinces. The higher genetic variation was found among landraces from Liaoning, followed by Anhui and Jiangxi,Mongolia was the lowest, [ Conclusion] The study indicates that ISSR markers produced high polymorphism in common buckwheat. ISSR analysis can be used for the molecular evaluation and studies of genetic diversity of buckwheat germplasm.
