化学损伤对成年大鼠骨骼肌肌卫星细胞的激活作用

摘  要:目的探讨激活骨骼肌肌卫星细胞的方法,提高肌卫星细胞的产量。方法取成年Wistar大鼠18只,随机分为6组,给大鼠下肢腓肠肌注射心脏毒素,分别于注射心脏毒素后的当天、注射后1、2、3、4、5d提取骨骼肌肌卫星细胞并计数,采用免疫荧光方法检测骨骼肌肌卫星细胞的纯度。结果采用Percoll分离液提纯细胞,纯度能够达到80%。注射心脏毒素后3、4d骨骼肌肌卫星细胞数最多,与其他时间比较,差异有显著性(F=5.540,q=4.576~5.829,P<0.05)。之后,细胞数开始减少。结论对骨骼肌进行一定程度的损伤,可以激活骨骼肌肌卫星细胞,从而提高肌卫星细胞的产量。Obective To explore the ways of activating and increasing the production of skeletal muscle satellite cells. Methods Heart toxin was injected into rat gastrocnemius muscle. The skeletal muscle were processed and the number of satellite cells counted the same day, and 1, 2, 3, 4, and 5 days after the injection. Immunofluorescence technique was used to detect the purity of the cells. Results By incontinuous density Pereol gradient centrifugation technique, the purity of processed cells reached 80%. The ceils were activated gradually after injection. The numbers of the ceils 3 or 4 days after injection were significantly higher than the other time-point groups (F=5. 540,q=4. 576-5. 829,P〈0.05). Conclusion A certain degree of injury to skeletal muscle can stimulate proliferation of skeletal muscle satellite cells.
