摘 要:目的探讨久效磷对小鼠的遗传毒性。方法50只小鼠,随机分成5组,设4个久效磷染毒组(0.25 mg.kg-1、0.50 mg.kg-1、1.00 mg.kg-1和2.00 mg.kg-1)和对照组。4个染毒组分别给予0.25 mg.kg-1、0.50 mg.kg-1、1.00 mg.kg-1和2.00 mg.kg-1的久效磷灌胃,对照组给予生理盐水灌胃,每日1次,连续14 d。采用微核试验和单细胞凝胶电泳实验检测久效磷的遗传毒性。结果久效磷染毒小鼠14 d后,与对照组相比,随着染毒浓度的升高,小鼠骨髓细胞微核率显著升高(P<0.05,P<0.01)),小鼠外周血淋巴细胞DNA损伤程度显著增加(P<0.05,P<0.01),并具有良好的剂量-效应关系。结论久效磷对小鼠具有遗传毒性。Objective To explore the genotoxicity of monocrotophos on mice.Methods Fifty mice were randomly divided into five groups.The experimental groups were given monocrotophos by gavage at several doses of 0.00,0.25,0.50,1.00 and 2.00 mg·kg^-1 respectively,one time a day for fourteen days.The control group was given saline by intragastric administration.Bone marrow micronuclei test and single cell gel electrophoresis were applied to test the genotoxicity.Results Compared with the control group,an increase in the rate of micronuclei and the DNA damage of peripheral lymphocytes in experimental groups were found(P〈0.05,P〈0.01).As to the rate of micronuclei and DNA damage,an obvious dose-effect relationship was showed.Conclusion Monocrotophos has genotoxic effect on mice.
