摘  要:利用PCR技术从天祝白牦牛基因组DNA中获得乳铁蛋白素(Lactoferricin,Lfcin)基因序列,将Lfcin基因连接于pGEM-T easy载体进行测序后,与奶牛的Lfcin基因序列进行比对;对牦牛、奶牛、人、小鼠等物种的Lfcin蛋白序列进行比对和进化树分析.结果表明:克隆获得了含天祝白牦牛乳铁蛋白第二外显子的DNA序列,共778 bp,其中Lfcin基因编码区长75 bp,编码25个氨基酸;序列分析显示,克隆获得的牦牛DNA序列与奶牛该序列存在9个碱基的变异,其中Lfcin基因编码区内有1个变异,为同义突变;牦牛和奶牛的Lfcin蛋白质序列完全相同,各物种Lfcin蛋白具有较高的同源性.Lactoferricin gene was obtained from genome of Tianzhu White yak by using PCR technology and cloned to pGEM-T easy vector, and the product was sequenced by bio-company. Tianzhu White yak and cow's Lfcin sequences were aligned; Yak, cow, human, mouse's Lfcin protein sequences were analyzed and their phylogenetic tree was established. The DNA sequence that contains the LF gene second exon of Tianzhu White yak was cloned, constituted by 778 bp, Lfcin coding region that codes 25 amino acids being 75 bp in length. According sequence analysis, there were 9 different bases between cow and yak's sequence; Yak and cow's Lfcin gene coded same protein sequence and there were high homology in different species. The phylogenetic tree of different species Lfcin protein accords with the order of evolution.
