摘 要:背景:碳纤维增强聚醚醚酮是聚醚醚酮的改进复合材料,改善了纯聚醚醚酮强度不足的缺点,扩大了其在各方面的使用范围。但若要成为应用于人体内的医用材料,必须具有良好的血液相容性。目的:比较碳纤维增强聚醚醚酮和钛合金对人血细胞的影响,判断碳纤维增强聚醚醚酮是否具有良好的血液相容性。设计、时间及地点:对比观察实验,于2008-06/08在吉林大学第三临床医学院中心实验室完成。材料:取实心钛合金股骨干髓内针1根及含20%碳纤维的碳纤维增强聚醚醚酮样条若干,机加工成棒形试样(长5mm,直径1mm)各20个。方法:在相同条件下,将钛合金和碳纤维增强聚醚醚酮2种材质的试件分别放置于新鲜人血中,通过观察溶血率,血小板激活程度以及白细胞激活程度判断材料的血液相容性。主要观察指标:应用游离血红蛋白直接测定法测量材料溶血率;流式细胞术测量血小板CD62p、CD63含量和白细胞表面黏附分子CD11b/CD18表达。结果:2种材料溶血率,血小板CD62p含量、CD63含量、白细胞表面黏附分子CD11b/CD18表达水平差异无显著性意义(P>0.05)。结论:碳纤维增强聚醚醚酮和钛合金对人血红细胞、白细胞和血小板无显著影响,均具有良好的血液相容性。BACKGROUND: Carbon fibers reinforced polyetheretherketone (CFR/PEEK) is a modified compound material of poly ether-ether-ketone (PEEK), improving weak intensity of pure PEEK and enlarging its serviceable range. However, excellent blood compatibility is necessary for clinical application. OBJECTIVE: To compare the effects of CFR/PEEK and titanium a/toy on human hemocytes, and determine whether CFR/PEEK has good blood compatibility. DESIGN, TIME AND SETTING: A contrast observational study was performed at the Central Laboratory of the Third Clinical Medical College of Jilin University between June and August 2008. MATERIALS: Machine-processed 40 stick samples (5 mm in length and 1 mm in diameter) were made using one titanium alloy femur stem nail and several CFR/PEEK samples (containing 20% carbon fibre) as raw materials. METHODS: Under the same conditions, the samples of titanium alloy and CFR/PEEK were placed in the fresh human blood so as to determine blood compatibility via haemolysis rate, platelet activation level, and leukocyte activation level. MAIN OUTCOME MEASURES: Haemolysis rate was measured with direct free hemoglobin assay; contents of CD62p and CD63 as well as CD11b/CD18 expression were measured using flow cytometry. RESULTS: There were no significant differences in heamolysis rate, contents of CD62p and CD63, and CD1 lb/CD18 expression between CFR/PEEK and titanium alloy (P 〉 0.05). CONCLUSION: Both CFR/PEEK and titanium alloy have no significant effects on human erythrocytes, leucocytes, and platelets, suggesting that they have a satisfactory blood compatibility.
