摘  要:目的研究液体深层培养猪苓菌丝体水提部位粗多糖脱色的最佳工艺条件。方法采用单因素试验和正交试验分别考察双氧水、活性炭、DEAE纤维素等3种脱色剂对菌丝体粗多糖的脱色效果,用光谱扫描法检测光吸收强度的变化。结果活性炭作为脱色剂,效果优于其他2种,脱色最佳工艺条件为:温度60℃,时间40min,活性炭添加量3g·100mL-1。结论实验中优化后的脱色方法简便、易行,也为猪苓菌丝体多糖的工业化提取中脱色工序的操作提供理论依据。OBJECTIVE To study the optimal technological conditions ofdecolorization of water-soluble crude polysaccharide from mycelium of Polyporus umbellatus of surbmerged fermentation. METHODS Water-soluble polysaccharide from the mycelium of fermented Polyporus umbellatus, was precipitated by alcohol and then decolored by H202, DEAE fibrin and activated carbon respectively. The decoloration efficiency was evaluated by light-absorption intensity using monofactorial and orthogonal testing. RESULTS The active carbon showed the priority in removing colored impurity effectively. Furthermore, three factors of affecting decolorization process were studied through orthogonal experiments of L9(3^4).The optimal technological conditions of decolorization were decided:solution temperature being at 60℃, process time being 40 min, and the amount of active carbon to the quantity of the polysarccharides solution being 3%. CONCLUSION The optimized method of decolorization, which can be easily operated, also supplies the theories foundation of decolorization technics in the commercial extraction process of polysaccharide from mycelium of Polyporus umbellatus.
