摘  要:目的研究缺血再灌注大鼠脑匀浆上清对干细胞的诱导作用。方法采用全骨髓培养法从SD大鼠骨髓中分离培养骨髓间充质干细胞(BMSCs),经贴壁法体外扩增、传代和鉴定。采用化学试剂和缺血再灌注后的大鼠脑匀浆上清分别诱导BMSCs培养物,免疫细胞化学染色检测其诱导后神经元特异性烯醇化酶(NSE)和胶质纤维酸性蛋白(GFAP)的表达。结果诱导后的BMSCs可表达CD44,不表达CD34。缺血再灌注后的大鼠脑匀浆上清诱导BMSCs分化的神经细胞更稳定,中等程度表达NSE和GFAP。结论在脑缺血再灌注后的环境下更有利于BM-SCs的神经分化。Objective To study the inducing effect of supernatant of brain homogenate in Ischemia-reperfusion rat on stem cell. Methods The SD rat bone marrow stem cells were isolated and cultured using the method of whole bone marrow culture, after ex vivo expansion, passaging and identified by CD44 and CD34, the BMSCs were induced by chemical reagent and supernatant of brain homogenate in Ischemia-reperfusion rat, respectively, then the expression of Neuron-Specific Enolase(NES) and Glial Fibrillary Acidic Protein(GFAP) were detected by immunocytochemical method. Results The expression of CD44 was positive and CD34 was negative in BMSCs, neurons induced by supernatant of brain homogenate in Ischemia-reperfusion rat were more stable, and showed middle degree expression of NSE and GFAP. Conelusiorts The environment of Ischemiareperfusion is more helpful to neural differentiation.
