摘 要:目的建立可用于临床样本检测的基于磁珠的可视化基因芯片检测方法。方法在玻璃芯片上以链霉亲和素包被的磁珠作为标记物,采用生物素标记的核酸为样品,通过链霉亲和素与生物素的亲和作用,进行核酸杂交检测,杂交结果以肉眼或通过普通显微镜观测,验证方法的特异性、灵敏性及重复性。结果方法检测的灵敏度为50ng,特异性和重复性均较好;对20份临床粪便样本分别进行PCR和芯片检测,芯片检测结果与PCR方法检测结果一致。20份临床样本中,6份阳性,其中1份为诺如病毒Ⅰ型、5份为诺如病毒Ⅱ型,14份阴性。结论方法显示具有高效、实用和可视化的特点,可同时检测样品中含有的诺如病毒Ⅰ型、诺如病毒Ⅱ型;检测时间短,能够满足口岸诺如病毒的快速检测要求,具有较好的实用价值,对于进一步开发更多指标的微生物检测方法具有很好的示范作用。Objective To develop a visual genechip with magnetic beads for detection of norovirus. Methods In the glass chip, streptavidin-coated magnetic beads were used as markers and biotinylated ohgonucleotides as sample. The results of the tests were analyzed. Results The sensitivity of the method was 50ng. The results of o 20 streptavidin and biotin samples were observed by the naked eyes or by an ordinary microscope. There 6 fecal samples were positive for norovirus, in which one sample belonged to norovirus type Ⅰ , five samples belonged to norovirus type Ⅱ .The other 14 samples were negative. Conclusion This method is efficient, practical and visual. It could be used to detect samples containing the norovirus type Ⅰ and Ⅱ, it can meet the requirements of the customs for rapid detection of norovirust.
