小儿抗痫胶囊对癫痫大鼠脑内GABA-T活性影响的实验研究

摘　　要：[目的]观察中药小儿抗痫胶囊对癫痫大鼠戊四唑点燃模型的影响,进一步从调节γ-氨基丁酸(GABA)代谢的角度探讨小儿抗痫胶囊的作用机制。[方法]动物模型采用大鼠戊四唑点燃模型。采用微量紫外比色法,观察大鼠大脑皮质、双侧海马γ-氨基丁酸转氨酶(GABA-T)活性的变化。[结果]中药组、西药组、模型组体质量与正常对照组相比有显著性差异(P<0.01),模型组体质量显著低于正常组,中药组较西药组体质量显著升高(P<0.01);模型组较正常对照组大鼠脑内GABA-T活性显著升高(P<0.01),中药组显著低于模型组(P<0.01)。[结论]抗痫胶囊对大鼠戊四唑点燃发作有显著对抗作用,其作用机制可能与其降低病模大鼠脑内GABA-T活性、谷氨酸(GLU)含量及增加GABA含量,影响脑内GABA代谢有关。[Objective] On the basis of past researches in the traditional Chinese medicine the effect of children anti-epilepsy capsule was observed in pentylenetetrazol （PTZ）-kindled model of rats and the mechanism of anti-epilepsy capsule was investigated at an angle of regulating metabolism of γ-aminobutiric acid （GABA）. [Method] The model of pentylenetetrazol （PTZ）-kindled epilepsy was used in the experiment. The activity of γ-aminobutiric acid -transaminase in homoginized brain tissue of the rats was measured with microuhraviolet spectrophotometry. [Results] The body weight of the antiepilepsy capsule group, sodium valproate group and the PTZ model group were significantly different when compared with the control group （P〈0.01）. significantly different compared with that of the The weight of the anti-epilepsy capsule group was PTZ model group （P〈0.01）. the activity of γ- aminobutiric acid -transaminase in the brain of model group was significantly higher than that in normal group（P〈0.01）. The activity in group treated with Chinese medicine was obviously lower than that in model group（P〈0.01）. [Conclusion] The mechanism of anti-epilepsy capsule on epilepsy may be related to decreasing the GABA-transaminase activity and the level of Glu in the cortex and hippocampus of the rats and to increasing the level of the GABA in the cortex and hippocampus of the rats.
