摘 要:目的和方法:实验以培养的人脐静脉内皮细胞(HUVEC)为研究对象,应用大剂量肿瘤坏死因子(TNF)造成HUVEC损伤,观察培养上清液血管紧张素转换酶(ACE)及HUVEC的超微结构变化,研究蜕皮甾酮(EDS)对内皮细胞损伤的影响。结果:大剂量TNF作用于HUVEC时,其培养上清液的ACE非常明显增高,超微结构破坏;应用EDS,则培养上清液的ACE有所减少、超微结构的损害程度有所减轻。结论:EDS能够减轻人血管内皮细胞的损伤,揭示了EDS在抗血管内皮细胞损伤方面的价值。AIM: To study effects of ecdysterone(EDS) on injuried vascular endothelial cells. METHODS:The vascular endothelial cells were damaged by recombinant human tumor necrosis factor α (rh-TNFα).The experiments were designed to observe changes of angiotensin converting enzyme(ACE) in culture liquid of human umbilical vein endothelial cells (HUVEC) and changes of ultrastructure of HUVEC.RESULTS: ACE activity in culture liquid of HUVEC distinctly raised and ultrastructure of HUVEC were destroied when HUVEC were injuried by a large dosage of rh-TNFα,but when EDS were applied in injuried HUVEC, ACE activity in culture liquid of HUVEC reduced and the damages of ultrastructure of HUVEC allevated. CONCLUSION:EDS was able to attenuate the injury of human vascular endothelial cells . This work revealed the value of EDS on treatment of injuried vascular endothelial cells. .
