摘 要:以TCM199为基础培养基,HMG(尿促性素)为激素,研究了HMG对猪卵母细胞体外成熟(IVM)和发育的影响;同时对EGF(表皮生长因子)和TGF-α(转化生长因子-α)单独或联合使用对猪卵母细胞IVM及发育的影响进行了研究,以确立较好的猪卵母细胞IVM培养体系。结果表明:(1)在猪卵母细胞IVM中分别添加0.075IU/ml、0.150 IU/ml、0.200 IU/ml、0.300 IU/ml的HMG,其中添加0.200 IU/ml的HMG组卵母细胞的成熟率和激活后的卵裂率显著高于其它组(P<0.05);(2)添加50 ng/ml EGF组成熟率和卵裂率显著高于对照组和添加30ng/ml、40 ng/ml、60 ng/ml组(P<0.05);(3)添加15 ng/ml TGF-α组成熟率显著高于对照组和添加5 ng/ml、40ng/ml组(P<0.05),激活后添加15 ng/ml组卵裂率显著高于其他组(P<0.05),当TGF-α的浓度高于15 ng/ml后对卵母细胞的成熟和激活后的卵裂没有显著的提高作用(P>0.05);(4)同时添加EGF和TGF-α组成熟率和卵裂率与对照组相比差异显著(P<0.05),但与添加50 ng/ml EGF、15 ng/ml TGF-α组相比没有明显差异(P>0.05)。可见EGF和TGF-α对猪卵母细胞的IVM和激活后的卵裂没有明显的协同作用。TCM199 was used as culture medium to discuss the effect of HMG as hormone on porcine oocytes maturation in vitro and development. At the same time effects of EGF, TGF-α separation or combination on porcine oocytes IVM and development were studied to establish better culture system. The results showed that : ( 1 ) The maturation rate and the cleavage rate derived from oocytes matured in medium supplemented with 0. 200 IU/ml HMG were significantly higher compared with 0. 075 IU/ml,0. 150 IU/ml,0. 300 IU/ml HMG groups (P 〈 0. 05 ) ; (2)The maturation rate and the cleavage rate derived from oocytes matured in medium supplemented with 50 ng/ml EGF were significantly higher compared with control and 30 ng/ml,40 ng/ml, 60 ng/ml groups (P 〈 0. 05 ) ; (3)The maturation rate derived from oocytes matured in medium supplemented with 15 ng/ml TGF-α was significantly higher compared with 5 ng/ml and 40 ng/ml groups (P 〈 0. 05). The cleavage rate was significantly higher with 15 ng/ml TGF-α than other groups( P 〈 0. 05 ). The maturation rate and the cleavage rate were not highter when the concentration of TGF-α was above 15 ng/ml; (4) The maturation rate and the cleavage rate derived from oocytes matured in medium supplemented with EGF and TGF-α were significantly higher compared with control( P 〈 0. 05 ), but it was not significantly difference with 50 ng/ml EGF and 15 ng/ml TGF-α (P 〉 0. 05). It showed that EGF and TGF-α did not have cooperative action.
