摘 要:目的评估转化生长因子β1(TGF-β1)在哮喘中调控上皮细胞修复的重要作用。方法通过将哮喘患者的上皮细胞与健康志愿者的上皮细胞相比较,评估TGF-β1对于表皮生长因子(EGF)诱导的增殖及其下游信号的影响。用溴代脱氧尿嘧啶核苷掺入法检测细胞增殖。结果通过将哮喘患者与健康志愿者的上皮细胞相比较,发现上皮细胞的基础增殖和EGF刺激后的增殖都明显减少,而且,ERK1/2磷酸化也减少了,这与EGFR磷酸化的减少相一致。TGF-β1的增加明显减少了EGF诱导的细胞增殖。哮喘细胞与正常细胞相比含有更多的TGF-β1。结论提示在支气管上皮细胞中,TGF-β抑制了EGFR磷酸化及其下游信号。Objective To evaluate transforming growth factor TGF-β1 TGF-β1 in regulating epithelial cell repair in asthma. Methods We evaluated the effect of TGF-β1 on epidermal growth factor (EGF)-induced proliferation and downstream signaling in epithelial cells obtained from subjects with asthma compared with cells from healthy subjects. Cell proliferation was evaluated by bromodeoxyuridine incorporation. Results Proliferation of epithelial cells at baseline and after EGF stimulation was significantly reduced in cells derived from subjects with asthma compared with cells obtained from healthy control subjects. EGF-induced ERK1/2 phosphorylation was reduced in epithelial cells from subjects with asthma compared with cells from healthy control subjects. This was paralleled with a reduced EGFR phosphorylation. Addition of TGF-β1 significantly decreased EGF-induced cell proliferation. TGF-β1 production was higher in asthmatic epithelial cells compared with normal cells. Conclusion Our results suggest that TGF-β restrains EGFR phosphorylation and downstream signaling in bronchial epithelial cells.
