摘 要:目的:探讨桔梗归肺经(肺靶向作用)的现代生物学机制。方法:观察桔梗对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)大鼠脑、肠、心、肝、肾、肺等组织中肠三叶因子(trefoil factor family3,TFF3)mRNA基因表达的影响。将实验大鼠分为假手术组、COPD模型组和桔梗组,分别取脑、肠、心、肝、肾、肺等组织,用RT-PCR技术测定不同组织中肠三叶因子(TFF3)mRNA表达。结果:①桔梗组与假手术组对照,脑、肠、肾、肺组织中TFF3mRNA基因表达虽有不同程度的升高,但无统计学意义;②桔梗组与模型组比较,脑、肠、心、肝、肾组织中TFF3mRNA基因表达有降低趋势,但无统计学意义;而肺组织TFF3mRNA基因表达显著上升,并具有统计学意义。结论:桔梗归肺经的作用机制,可能与其能够增加疾病状态下肺组织中TFF3mRNA基因表达有关。Objective: To explore modem biological mechanism of meridian tropism of Radix Platycodi (target effect). Methods: Observe the influence of Radix Platycodi on TFF3 gene expression on the tissue of brain,intestine, heart,liver, kidney, lung in COPD rats. Rats were divided into sham operated group, model group and Radix Platycodi group. Measure TFF3 gene expression on brain, intestine, heart, liver, kidney, lung by RT-PCR method. Results; (1)TFF3mRNA expression on the tissue of brain, intestines, kidney, lung were higher in Radix Platyeodi group than that in sham operated group, but have no statistic difference.(2) TFF3mRNA expression of brain, intestines, heart, liver, kidney show a degressive trend in Radix Platycodi group compare with that in model group, but have no statistic difference. TFF3mRNA expression on lung tissue increase significantly,. Conclusion : The mechanism of meridian tropism of Radix Platycodi may be related to the increase of TFF3mRNA expression on lung tissue in disease condition.
