摘  要:目的:探讨醋酸铅对体外培养未成年小鼠骨髓间充质干细胞生长的影响,为进一步阐明铅骨髓毒性作用机制,采取相应防治措施提高儿童健康水平提供依据。方法:不同浓度醋酸铅作用于体外培养的骨髓间充质干细胞,观察其对间充质干细胞成集落(CFU-F)数的影响及变化规律。结果:醋酸铅可明显抑制CFU-F的生长,各实验组与对照组比较差异有统计学意义(P<0.05),且随浓度增加抑制作用增强,呈明显的剂量-效应关系(r=-0.960,P=0.000),其中CFU-F集落的IC50=80.00μmol/L。结论:醋酸铅可明显抑制体外培养的骨髓间充质干细胞生长。Objective: To explore the effect of lead acetate on the growth of immature murine mesenchymal stem cells in vitro. Methods : Lead acetate of different concentrations were used in the culture of colony - forming unit - fibroblast ( CFU - F) , the effect on the colony- forming and therule of variation were observed. Results: CFU -F colony rate reduced in a dose -dependent depression of the colony numbers ( r = - 0. 960, P = 0. 000 ) . Meanwhile, compared with the control group, the differences of every experiment groups were remarkable ( P 〈 0. 05 ). The ICS0 of CFU - F colony was 80.00μmol/L. Conclusion: The data suggests CFU - F reduces when poisoned by lead acetate obviously in vitro.
