摘 要:目的研究肌肽对缺氧所致人体内皮细胞系ECV304细胞损伤的影响。方法建立缺氧条件下ECV304细胞损伤模型,用MTT法观察肌肽对缺氧损伤的ECV304细胞活性的影响,测定细胞培养基中乳酸脱氨酶(LDH)活力,并对细胞骨架进行考马斯亮蓝R-250染色观测其细胞结构。结果浓度为10~20 mmol/L肌肽孵育ECV304细胞6 h后,可以抑制缺氧12 h和24 h引起的ECV304细胞活性下降,同时减少LDH的释放,保持细胞骨架完整。结论肌肽对缺氧所致的ECV304细胞伤具有保护作用。Objective To investigate the effect of camosine on the injury of human umbilical vein vascular endothehal cell line ECV304 induced by hypoxia. Methods The model of the injury of ECV304 cells induced by hypoxia was established. The effect of carnosine on the activity of ECV304 cells was determined by MaT assay. The level of lactate dehydrogenase ( LDH ) in the cell medium was measured with the corresponding kit. The cell structure was observed under microscope after Coomassie brilliant blue R-250 staining. Results After culturing ECV304 cells with carnosine ( 10 to 20 mmol/L ) for 6 hours,the decrease in ECV304 activity induced by 12- and 24-hour hypoxia was inhibited. The release of IDH was also inhibited,and the integrity of cell structure reamained. Conclusion Camosine has protective efiect on the injury of ECV304 induced by hypoxia.
