摘 要:目的培养大鼠心脏微血管内皮细胞,探讨其对研究血管病变的意义。方法采用Nishida的方法,在无菌条件下分离Wistar大鼠心脏,用胶原酶、胰酶进行消化,通过离心,收集细胞进行培养。结果培养的细胞呈单层贴壁生长,细胞初为圆形,贴壁后为短梭形,5~7 d呈"铺路石样"生长,培养的细胞在人工基膜(Matrigel)上形成管腔样结构。Ⅷ因子相关抗原免疫细胞化学鉴定为阳性。结论本方法简单易行,对血管病变的研究有重要意义。Objective To culture, micmvascular endothelial cells from rat heart,and to assess its significance in vascular pathological changes. Methods According to Nishida method,the microvascular endothelial cells were isolate from Wistar rat hearts after sequence di- gestion with collagenase and trypsin, separated by differential centfugation, and then cultured. Results After 5 to 7 days, the cultured cells displayed monolayer growth,looked like irregular cobblestone road, and formed capillary-like tube on Matrigel. The factor Ⅷ-related antigen was positive in immunohistochemical assay. Conclusion This study provides a simple and easy method to culture microvascalar endothelial cells from rat heart in vitro, which is of great value for studying the vascular pathological changes.
