摘  要:目的观察短期禁食对大鼠心肌缺血再灌注后心肌细胞、线粒体的结构和功能的保护作用。方法40只SD大鼠,随机分为对照组、缺血再灌注组(I/R)、短期禁食组(STF)、缺血预处理组(IP)、短期禁食+缺血预处理组(STF+IP)。建立I/R实验模型。用分光光度计检测线粒体丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSHPx)活性;透射电镜观察心肌超微结构的改变。结果与对照组相比,I/R组MDA含量明显升高,SOD及GSHPx活性明显降低;STF和IP组MDA升高,SOD、GSHPx降低不如I/R组明显。I/R组心肌细胞及线粒体损伤重,STF及IP组心肌超微结构破坏减轻,STF及IP联合应用对心肌细胞的保护作用更明显。结论短期禁食通过减轻心肌细胞和线粒体损伤对在体大鼠心肌缺血再灌注损伤具有一定的保护作用。Objective To observe the effects of short-term fasting on myocardial mitochondria after myocardial ischetnia/reperfusion in rats. Methods 40 SD rats were randomly divided into 5 groups:normal control group,ischemic/reperfusion group(I/R),short-term fasting group (STF), ischemic preconditioning group (IP), short-term fasting and ischemic preconditioning group(STF+IP). The I/R model of SD mrs was established. The level of MDA,SOD and GSHPx of myocardial mitochondria were detected by spectrophotometer. Uhrastructure changes of myocardium were observed under electronic microscope. Results Compared with control group, the content of MDA in group I/R was increased, and SOD and GSHPx activities were decreased. The changes of MDA content, SOD and GSHPx activities in groups STF and IP were less than those of group I/R. Compared with groups STF and IP,there was a significant difference in contents of MDA,SOD and GSHPx in group STF+IP. There is no significant difference between groups STF and IP. Ultrastructural injuries of myocardial mitochondria in groups STF and IP were milder than those in group I/R. Conclusion Short-term fasting can attenuate the changes in the contents of MDA, SOD and GSHPx induced by I/R and alleviate myecardial I/R injury.
