摘 要:目的观察华蟾素与紫杉醇配伍对小鼠S180腹水瘤和肉癌移植瘤的作用。方法ICR小鼠分别于腹腔和腋下接种1×106个S180肿瘤细胞,腹腔注射华蟾素注射液(80,20,5mg.kg-1)或(80,20,5mg.kg-1)同时腹腔注射紫杉醇注射液5mg.kg,连用9天。腹水瘤模型连续给药9d后停药,观察小鼠的生存时间,计算荷瘤小鼠生命延长率。肉癌模型连续给药9d后,次日取瘤块称重,计算肿瘤抑制百分率。结果在腹水瘤模型中,与生理盐水对照组的(17.3±1.3)d比较,华蟾素高,中,低剂量单用组小鼠存活时间分别为(21.5±2.4)d(P<0.05),(18.6±1.4)d,(18.3±2.4)d;合用组:与生理盐水组比较均有显著性差异,平均小鼠存活时间分别为(26.6±1.6)d(P<0.001),(24.3±2.1)d(P<0.001),(21.8±1.1)d(P<0.01);对腹水瘤小鼠的生命延长率分别是56.4%,40.4%和26.0%。肉癌模型中,与生理盐水对照组的(1.55±0.52)g比较,单用组:S180癌平均瘤重分别为(1.05±0.48)g(P<0.01),(1.44±0.41)g,(1.39±0.60)g;合用组:与生理盐水组比较均有显著性差异,平均瘤重分别为(0.61±0.40)(P<0.001),(0.88±0.38)(P<0.001),(1.09±0.55)(P<0.01)g;抑瘤率分别为60.7%,43.3%和30.0%。〗结论华蟾素与紫杉醇合用有明显的协同治疗作用。OBJECTIVE To study on the effects of Cinobufacin combined with taxol injection on S180 tumor in vivo. METHODS Tumors were established by injection of S180 cells(1 × 10^6 cells per animal, to form S180 ascites or subcutaneously solid tumor in ICR male mice).Cinobufaein (80, 20, 5mg·kg^-1) and it combined withtaxol(80, 20, 5mg·kg^-1 ± 5mg·kg^-1, respectively) were administrated ip for consecutive 9 days. In mice bearing S180 ascites, the prolongation rate of life were recorded. In mice bearing S180 sarcoma, mice were sacrificed at day 10 and tumor weight were recorded. RESULTS In mice bearing S180 aseites, the survival times of Cinobufaein groups were (21.5 ± 2.4) days (P 〈 0.05), ( 18.6 -± 1.4) days, ( 18.3 ± 2.4) days, compared with the control (normal saline)group in which the survival times was (17.3 -± 1.3)days. The survival times of combined groups were (26.6± 1.6)days (P〈0.001), (24.3± 2.1)days (P〈0.001), (21.8 ± 1.1)days (P〈0.05).The prolongation rate of life were 56.4 %, 40.4 % and 26.0 % respectively. The tumor weight of Cinobufacin group were (1.05 ± 0.48)g(P〈0.01), (1.44 ± 0.41)g, (1.39 ± 0.60)g, compared with the control(normal saline)group in which the tumor weight was( 1.55 ± 0.52) g. The tumor weight of combined groups were(0.61 ± 0.40), (0.88 ± 0.38), (1.09 ± 0.55)g, and the tumor inhibition rate were 60.7 %, 43.3 % and 30.0 respectively. Conclusion Cinobufaein combined with taxol could inhibit the proliferation of cancer cells efficiently.
