摘  要:目的评价白藜芦醇(Res)的抗癫痫效果并初步分析其作用机制,为开发新型抗癫痫药物提供理论依据。方法建立颞叶癫痫大鼠模型后,以Res干预10d,丙戊酸钠(VPA)作为阳性对照,观察Res对颞叶癫痫大鼠的行为学、电生理学和病理学指标的影响。结果模型组自发性发作率增加,海马内神经元大量死亡,苔藓纤维出芽(MFS)至颗粒细胞层和内分子层;Res能抑制癫痫大鼠自发性发作,对并海马CA1和CA3a区神经元有保护作用和抑制MFS(P<0.05)。结论Res具有较强的抗癫痫作用。Objective To evaluate the possible anti-epileptic effect of resveratrol(Res) against kainic acid (KA)- induced temporal lobe epilepsy (TLE) in rats. Methods TLE was induced by KA (1μg) injected into the center of the CA3 region of the right hippocampus with stereotaxie technology. The vehicle solution, Res, and valproate (VPA) were administered intragastrieally immediately following the first onset of seizure in acute stage, once per day, for ten days. After intragastric administration of the agents, behavior monitoring, intracranial electroence- pholography recording, and histological analysis were performed to evaluate the anti-epileptic effect of Res. Results The rates of spontaneous seizures in epileptic rats increased. The Res decreased the frequency of spontaneous seizures in rats, inhibited the epileptiform discharges, protected neurons in CA1 and CA3a regions, and depressed mossy fiber sprouting, which are general histological characteristics both in TLE patients and animal models. Con- clusion Our study suggests that Res is a potent anti-epilepsy agent, and protects against epileptogenesis and pro- gression of the KA-induced TLE animal model.
