摘  要:目的考察体外溶血实验葛根素注射液浓度对红细胞膜脆性的影响及与红细胞溶血率的相关性。方法用热诱导法考察50℃时葛根素注射液对Beagle犬红细胞膜脆性的影响;将不同浓度的葛根素溶液分别与Beagle犬红细胞、豚鼠红细胞在温度为37℃的生化培养箱内孵育6 h,于不同时间点观察各样品管的溶血程度,并于6 h后通过紫外分光光度计测定各管在波长540 nm处的吸光率,计算溶血率。结果高温条件下葛根素注射液可加剧Beagle犬红细胞溶血程度;葛根素注射液均能直接引起Beagle犬、豚鼠红细胞溶血,溶血率与葛根素浓度成明显的量效关系。结论葛根素注射液在一定浓度下可引起红细胞膜脆性增加;随着浓度的升高葛根素注射液的溶血作用更明显。Objective To investigate the effect of the concentration of puerarin injection on the stability of animal red blood cells (RBC) . Methods Suitable Beagle dogs'RBC was added to the Puerarin Injection dilution at different concentrations in the glass the and then were incubated Incubate tubers at 50℃ for 30 min, and the absorbency of the liquid supernatant was tested and the hemolytic rate was calculated . The same operation as above was conducted when of RBC Guinea pigs was incubated at 37 ℃ for 6 hs. Results When incubating at 50℃ ,the hemolytic rate of Beagle dogs'RBC could be up to nearly 50% with high concentration of puerarin. The same situation in the tests at normal physical temperature, and the hemolytic rates of animals'RBC showed close relationships with the concentration of puerarin. Conclusion Puerarin Injection can destroy the stability of RBC. Either at high temperature or at normal physical temperature the dilution of Puerarin Injection can worsen dramatically the hemolysis of animal's RBC as long as the increasing concentration of puerarin.
