摘  要:目的探讨慢性支气管炎模型的造模方法。方法24只大鼠随机分为正常组(A组)、单通道脂多糖组(B组)、双通道脂多糖组(C组)。A、B组分别给予气管内一次性注入生理盐水和脂多糖200μg,C组在B组的基础上,第2天开始,连续4d经鼻滴入脂多糖50μg.200μl-1.d-1。3周后大鼠全部处死,观察大鼠肺支气管HE染色、AB-PAS染色的病理形态学改变,并进行半定量评分。光镜下行支气管肺泡灌洗液(BALF)计数和分类,免疫组化测定肺组织黏液蛋白Muc5ac的表达,并进行半定量评分。结果与A、B组相比较,C组大鼠肺气道上皮炎性细胞浸润、纤毛粘连脱落、黏膜充血水肿等病理表现明显(P<0.05),杯状细胞增生计数增加(P>0.05)。BALF白细胞总数及中性粒细胞百分比均升高(P<0.05),Muc5ac的表达亦明显增加(P<0.05)。结论双通道给脂多糖比单通道更能建立典型的慢性支气管炎大鼠模型。Objective To establish the chronic bronchitis model with double-channel tecnique in rats. Methods Twenty-four male Wistar rats were randomly divided into 3 groups of normal control group(A) ,mono-channel LPS group(B) and double-channel I.PS group(C) with 8 rats each.. Rats in group 13 were intratracheally instilled with lipopolysaccharide 0.2 mg, rats in group C with lipopolysaccharide 0.2 mg plus daily instillation of lipopolysaccharide 50 μg·200μl^-1· d^-1 via nose for consecutive 4 days. All rats were bred 3 weeks later. The morphological changes of airway were observed. Total and differential cell counts in bronchoalveolar tavage fluids(BALF) were measured. The expression of Muc5ac in lung tissues was detected by immunohistochemistry. Results Compaered with group A and B, there were the pathological features of goblet cells such as invantion of inflammatory cells in group C (P〈0.05). The number of cells and the percentage of neutrophils in BALF in group C were significantly greater than those in group A and B (P〈0. 05). The expression of Muc5ac was higher in group C than that in group A and B (P〈0.05). Conlusion The doublechannel tecnique is better than mono-channel model in establishing a rat chronic bronchitis model with typical pathological features.
