摘 要:目的探讨迷走神经结状神经节与心肌细胞之间的联系与作用方式。方法体外联合培养心肌细胞与迷走神经结状神经节,采用相差显微镜观察两者联合培养不同时间的活细胞生长状况。结果联合培养24~48h,可见结状神经节组织块周围有神经元迁出,组织块长出的神经纤维呈放射状向四周延伸。联合培养72~96h,得到具有选择性结状神经节神经元支配的心神经纤维在单层分散的心肌细胞表面相互交织成网状,并可见有神经终末终止于分散培养的心肌细胞的表面。联合培养7d后,单纯培养心肌细胞逐渐停止搏动并变性、死亡,而联合培养中心肌细胞仍搏动良好。结论联合培养的心肌细胞诱导了结状神经节神经元的迁移,并且神经元与心肌细胞之间可能通过神经-肌肉接头的方式进行联系。Objective To investigate the relationship between nodose ganglion and cardiac muscle cells. Methods The neuromuscular co-cultures of nodose ganglion and cardiac myocytes were established. The living ceils of the cultures were observed with an inverted contrast microscope at different culture time. Results The nodose ganglion cells were observed with a lot of processes 48h after co-cultured with cardiac muscle cells, the processes formed a network on the surface of muscle cells 72h after co-culture, fibers terminated on the surface of muscle cells 96h after co-culture, some fiber endings bifurcated and terminated on cardiac muscle cells 78h after co-culture, some fiber endings terminated on cardiac muscle cells with fan-shape 96h after co-culture. The co-cultured cardiac muscle cells still functioned and beat well 7 days After co-culture, but the nonco-cuhured cardiac muscle cells stopped beating and degenerated and died. Conclusion The co-cultured cardiac myocytes have inductive effect on nodose ganglia through the neuromuscular junction.
