摘  要:目的研究严重糖尿病足坏疽血管组织中的差异表达基因及糖尿病坏疽的可能机制。方法应用功能分类基因芯片筛选糖尿病坏疽血管组织中的差异表达基因;用反转录多聚酶链反应(RT-PCR)对3个差异表达基因进行验证。结果在113个基因中,有13个基因的表达水平上调2倍以上,3个基因的表达水平下调2倍以上;RT-PCR检测的其中3个基因,与芯片检测的结果一致。结论涉及多种基因表达异常的血管组织病变在糖尿病坏疽的发生发展过程中占重要地位;分析差异基因的表达,有助于了解糖尿病坏疽发生的可能机制。Objective To study the differentially expressed genes in the vascular tissue of diabetic gangrene toot and the possible mechanism of diabetic gangrene. Methods The differentially expressed genes in the vascular tissue of diabetic gangrene loots were screened by the functional elassific gene chip. Reverse transcription polymerase chain reaction (RT-PCR) detection was used to verify the differentially expressed 3 genes. Results In the detected 113 genes, 13 of the gene expression level increased more than 2 times, 3 of the gene expression level decreased more than 2 times. Three genes were verified with RT-PCR detection, which were results similar to those with chip testing. Conclusions The abnormal expression in a variety of genes in tissues of vascular lesions plays an important role in the course of development and progression of diabetic gangrene. Analysis of differential gene expression can contribute to understanding of the possible mechanisms of diabetic gangrene.
