摘  要:目的探讨川芎嗪(LH)促进小鼠骨髓基质细胞(BMSC)生长及干预长春新碱(VCR)抑制小鼠BMSC增殖的作用。方法①LH5、10、20、40μg/mL,以及VCR2.5、5、10、15μg/mL分别与BMC共同培养14d,测定细胞增殖。②LH5、10、20、40μg/mL与BMC预培养1h后再分别加入终浓度为5μg/mLVCR继续培养14d,测定细胞增殖。结果LH20、40μg/mL组、VCR10、15μg/mL组OD值与对照组比较差异均有统计学意义(均P<0.05),LH20、40μg/mL组OD值与LH10μg/mL组比较差异均有统计学意义(均P<0.05);VCR2.5μg/mL组OD值与VCR15μg/mL组比较差异有统计学意义(P<0.05)。LH10μg/mL+VCR5μg/mL组、LH20μg/mL+VCR5μg/mL组、LH40μg/mL+VCR5μg/mL组OD值与VCR5μg/mL组比较差异均有统计学意义(均P<0.05);LH20μg/mL+VCR5μg/mL组、LH40μg/mL+VCR5μg/mL组OD值与对照组比较差异均有统计学意义(均P<0.05)。结论VCR明显抑制BMSC生长;LH可促进BMSC生长且具有干预VCR对BMSC增殖的抑制作用。Objective To explore the effect of ligustrazine hydrochloride(LH) for injection on the proliferation of bone marrow stromal cells(BMSC) inhibited mice by vineristine(VCR). Methods ①The marrow cells were cultured with LH (5,10,20,40 μg/mL) and VCR (2.5,5,10. 15 μg/mL) in 37 ℃ 5%CO2 for 14 days, respectively and their effect on marrow stromal cells proliferation was determined. ②The marrow cells were pretreated by LH(5.10.20,40 μg/mL) for 1 h,then cultured with VCR 5 μg/mL for 14 days,and the effect of LH on marrow stromal cells proliferation was determined. Results There was statistical signi tween LH groups of 20.40μg/mL,VCR groups of 10.15 ug /mL and ficance for OD value becontrol group(P〈0. 05, respectively). There was statistical significance for OD value between LH groups of 20.40μg/mL and LH group of 10 μg/mL (P〈0.05, respectively). There was statistical significance for OD value between VCR group of 2.5 μg/mL and VCR group of 15μg/mL(P〈0.05). There was statistical significance for OD value between groups of LH 10 ug /mL^-1 VCR 5 μg/mL, LH 20 μg/mL+VCR 5 μg/mL, LH 40 μg/mL+VCR 5μg/mL and group of VCR 5 μg/mL(all P〈0.05). There was statistical significance for OD value between groups of LH 20 μg/mL+VCR 5 μg/mL, LH 40 μg/mL+VCR 5 μg/mL and control group (P〈0.05, respectively). Conclusion The results suggest that vincristine could inhibit the BMSC proliferation. Ligustrazine could promote the BMSC growth and the proliferation of BMSC inhibited by vincristine.
