摘 要:五味子乙素(Schisandrin B,Sch B)是传统中药五味子的主要活性成分之一,文章利用体外原代培养的海马神经元网络作为实验模型,研究Sch B对神经元网络功能的调控作用。实验结果表明:Sch B对神经元网络自发同步钙振荡有显著抑制作用,且呈浓度依赖,IC50=1.61±0.11 mg/L。提示Sch B能够有效调节突触终末自发递质释放、调节神经元网络兴奋性、具有神经保护性。Schisandrin B (Sch B) is one of the main effective composites of Schisandra chinensis, a famous Chinese medicine, which is widely used as an anti-stress, anti-aging and neurological performance improving herb. In this study, the authors examined its specific effect of modulating calcium oscillation frequency in cultured hippocampal neural networks. The frequency of intracellular spon- taneous and synchronous calcium oscillations was depressed to 49. 0 ±8.0% after 3 - 4 min of Sch B (2. 0 mg/L) application. The inhibitory effect was dose dependent and IC50 = 1.61 ± 0. 11 mg/L. The results suggest that Sch B could modulate excitability of neuronal networks through effecting spontaneous neurotransmitter release in the synaptic terminals, and could be neuroprotective through inhibiting synchronous activities and intracellular calcium accumulation.
