摘 要:以60Co-γ射线辐照东方百合鳞片后组培诱导芽的发生及增殖,对增殖的不定芽进行诱导生根以探讨辐照处理对不定芽生根的影响及射线辐射的细胞学效应.结果表明:辐射处理对不定芽生根率的影响不明显,而对生根芽叶片数、根数、根长等各生长指标的影响均达到极显著水平;在0.5~4.0Gy范围内,再生植株芽高随辐射剂量升高呈显著下降的趋势.在0.5~1.0Gy范围内对根尖细胞的有丝分裂表现为刺激效应,在2.0~4.0Gy范围内表现为抑制效应;各辐照处理的畸形细胞率均高于对照,其中以2.0Gy处理最高;辐照处理对再生植株根尖细胞染色体畸变类型的影响程度不同,染色体桥率受辐射剂量影响最严重,其次是落后染色体率和染色体片段率,对微核率产生的影响最弱.不同形态变异再生植株在染色体水平上的变异情况不同.1.0Gy辐照处理是百合鳞片辐照结合组培的适宜辐照剂量.The oriental lily scales were irradiated with ^60Co-γ rays and cultured in vitro to induce bud and proliferate. The proliferation buds were induced to root and explore the effect of ^60Co-γ rays radiation on the plantlet growth and its cytological effect. The result shows that the effect of irradiation on the rooting rate was not significant, while its effects on the growth indexes of plantlets, such as number of leaves, number of roots, the root length were most significant. Within the treatments of 0.5-4.0 Gy radiation doses, the height of regenerating plant was declined with the dose increasing . The effect of irradiation on the karyokinesis of root tip cells of plantlets represented stimulating within the treatments of 0. 5- 1.0 Gy radiation doses and restraining within the treatments of 2. 0- 4.0 Gy. The cell aberration rate of all the irradiation treatments were higher than the control, the aberration rate of the 2.0 Gy treatment was the highest. Types of chromosome abnormity of root tip ceils were affected incoordinately by different doses of irradiation. The effect of irradiation on percent of chromosomal bridge was the most sensitive, then the percent of lag chromosome and chromosomal fragment, the percent of micronuclei aberration was the least sensitive. Chromosomal aberrance types of morphological abnormal shoots were different. 1.0 Gy was a fitting dose for the irradiation treatment combining with culture in vitro on the oriental lily scales.
