摘 要:目的:建立高效液相色谱法,同时测定茚地那韦和依非韦伦在人血浆中的药物浓度。方法:色谱柱为岛津Shim-packCLC-ODS(6mm×15cm,5μm),流动相为乙腈-磷酸盐缓冲液(50:50),流速1.2mL.min-1,紫外检测波长:0~10min为260nm,10.01~25min为240nm。柱温为35℃。内标地西泮与茚地那韦和依非韦伦的保留时间分别为9.824,5.454,18.010min。结果:茚地那韦和依非韦伦线性范围分别为0.05~20mg.L-1(r=0.9990)和0.1~10mg.L-1(r=0.9996),最低检测限分别为0.05mg.L-1和0.1mg.L-1,样品溶液冻融稳定,日内精密度和日间精密度良好,回收率较高。结论:采用HPLC同时测定茚地那韦和依非韦伦血浆药物浓度,方法简便,结果准确可靠。OBJECTIVE To establish a method for simultaneous determination of indinavir and efavirenz in human plasma. METHODS The concentrations of indinavir and efavirenz were analyzed on a Shim pack CLC-ODS column(6 mm× 15 cm, 5μm)with a mobile phase consisting of water-acetonitrile (50: 50) at a flow rate of 1.2 mL·min ^-1. The samples were determined with wavelength program. The temperature of column was at 35 ℃. The retention times of indinavir, internal standard (Diazepare) and efavirenz were 9. 824 rain, 5. 454 min and 18. 010 min, respectively. RESULTS The calibration curves were linear in the ranges of 0.05 - 20 mg·L ^-1 for indinavir (r= 0. 999 0)and 0. 1 - 10 mg·L ^-1 for efavirenz (r= 0. 999 6), respectively. The average recoveries of cryopreservation, free-thaw and precisions of indinavir and efavirenz were both well. CONCLUSION The method is simple, practical and reliable,which could be used for the study of pharmacokinetics and pharmacodynamics.
