摘  要:目的研究宝宝乐口服液对瘦素调节下丘脑腹内侧核(VMH)神经元膜电位的影响。方法利用红外可视脑片膜片钳技术记录对照组、厌食组和治疗组三组模型动物VMN神经元的膜电位,分析瘦素及宝宝乐口服液对VMN神经元膜电位的影响。结果瘦素使三组动物大部分神经元去极化,各组间比较差异无显著性。少数VMN神经元在瘦素作用下超极化,各组间比较差异亦无显著性。但模型组VMN神经元的膜电位被瘦素去极化而导致自发放电增加的细胞比例增多,被瘦素超极化而自发放电减少的细胞比例减少,对瘦素无反应的细胞比例也减少;而治疗组去极化、超极化和无反应的细胞数与对照组差异无显著性。结论宝宝乐口服液能够通过影响降低VMN对瘦素的整体敏感性,使VMN发出适度的饱信号,从而恢复正常的食欲和摄食量。Objective To study the effects of Baobaole Decoction on leptin-regulated membrane potential of VMN neuron. Methods By using infrared video patch-clamp, the membrane potential of three animal models which are the control groups, anorexia and treatment group were recoded and the effects of Baobaole Decoction and leptin on the membrane potential of YMN neuron were analyzed. Results Leptin depolarized the most neurons of the three model animal groups and hyperpolarized the minor neurons, but there had no significant difference among the three groups. However, leptin increased the proportion of the spontaneous discharge neurons with depolarizing MP of the VMN neuron in model group, but decreased the proportion of the spontaneous discharge neurons with hyperpolarizing membrane potential of the VMN neuron. But the cell numbers of depolarization, hyperpolarization and unresponse in the treatment group showed no significant difference. Conclusion In order to recover normal appetite, Yunpi prescription decreased the sensitivity of YMN neurons to leptin and made VMN neurons to send out moderate satisfied signal to induce anorexia.
