金丝桃苷对SLT-Ⅱe诱导大鼠肠黏膜微血管内皮细胞分泌功能的影响  被引量:1

摘  要:将肠黏膜微血管内皮细胞分为正常对照组、SLT-Ⅱe对照组及1、5、10μg/mL金丝桃苷处理组5组,采用ELISA法测定了培养3、6、9和12 h时细胞上清液中NO、ET-1、P-选凝素、sICAM-1及TNF-α浓度的变化。结果显示,5μg/mL金丝桃苷可下调SLT-Ⅱe诱导的大鼠肠黏膜微血管内皮细胞NO、ET-1的分泌,细胞培养至第12 h时,NO分泌量为(19.04±1.38)μmol/L,ET-1分泌量为(2.54±0.25)pg/mL。10μg/mL金丝桃苷可下调SLT-Ⅱe诱导的大鼠肠黏膜微血管内皮细胞sICAM-1的分泌,第12 h时其浓度为(9.81±1.92)pg/mL。1、5、10μg/mL金丝桃苷不能下调SLT-Ⅱe诱导的大鼠肠黏膜微血管内皮细胞P-选凝素和TNF-α的分泌。表明,金丝桃苷可通过抑制SLT-Ⅱe诱导肠黏膜微血管内皮细胞NO、ET-1、sICAM-1的过量分泌,缓解肠道微循环障碍,减轻炎症反应,达到治疗水肿病的目的。The cultured rat intestinal microvascular endothelial cells(RIMECs) in vitro were divided into control group,SLT-Ⅱe group, test Group 1 (1μg/mL SLT-Ⅱe +1μg/mL hyperoside), test Group 2(1 μg/mL SLT-Ⅱe +5μg/mL hyperoside) and test Group 3(1μg/mL SLT-Ⅱe +10μg/mL hyperoside), respectively. The concentration of NO, ET-1, P-selectin, sICAM-1 and TNF-α in cell supernatant were determined using ELISA at hours 3,6,9 and 12 post cultivation, respectively. The results showed that 5μg/mL hyperoside significantly reduced the secretion of NO and ET-1, and 10μg/mL hyperoside signifi-cantly reduced the secretion of sICAM-1. The secretions of NO, ET-1 and sICAM-1 were (19.04±1.38)μmol/L, (2.54±0.25) pg/mL and (9.81±1.92) pg/mL at hour 12 post-cultivation,respectively. The secretions of P-selectin and TNF-α were not reduced by 1μg/mL,5μg/mL and 10μg/mL hyperoside. It was concluded that hyperoside could reduce the secretions of NO, ET-1 and slCAM-1 from rIMECs to relive instestinal microcirculation obstruction,to alleviate the inflammation response and to treat edema disease of swine.
