山羊大腔卵泡卵母细胞的体外成熟和体外受精

摘　　要：从屠宰场采集卵巢。抽吸法回收2～6mm大腔卵泡卵母细胞。随机抽样后，测量其直径为167.42±17.43μp。A，B级卵泡卵母细胞进行体外成熟培养，其成熟率为67.50％。山羊新鲜精液以BO液或MD-H液处理后，与体外成熟卵母细胞进行授精，受精率分别为56.61％（17／30）和63.33％（19／30），两组间差异不显著（P＞0.05）。BO和MD-H液均适宜于山羊精子获能处理。体外受精卵的2细胞胚发育率为16.67％（6／36），4细胞胚发育率为2.78％（1／36）。Goat ovaries used in our research was collected from slaughter house. Ova were aspirated from the follicles of 2～6mm (in diameter) and its diameter were measured, and the result was 167. 42 ±17. 43μm. Selecting oocytes of grade A and B for IVM, the maturation rate was 67. 5 %. Fresh goat sperms were incubated in BO and MD - H media supplemented with 20mg/mL BSA and 10μg/mL Heparin for capacitation. Capacitated sperm was cocultured with matured oocytes of insemination. The fertilization rate was 56. 67 % (17/30) and 63. 33 % (19/30)respectively. It showed that there were no significant differences between the two dealted methods. The results indicated that both BO and MD - H media suited for goat spermatozoa capacitation. The rates of cleavage were 16. 67% (6/36).
