摘 要:目的观察商陆水煎剂对阿霉素肾小球硬化大鼠肾脏的保护作用。方法采用单侧肾切除并尾静脉重复注射阿霉素的方法建立大鼠肾小球硬化模型。将48只SD大鼠随机分为假手术组、模型组、商陆低剂量组、商陆高剂量组,分别给予生理盐水、生理盐水、商陆水煎剂(2g.kg-1.d-1)、商陆水煎剂(4g.kg-1.d-1)治疗,连续治疗93d。观察商陆水煎剂对肾小球硬化大鼠尿蛋白、血浆白蛋白、血肌酐(Scr)、尿素氮(BuN)和肾脏病理改变及肾小球硬化指数的影响。结果与模型组比较,术后第8、13周商陆高、低剂量组大鼠尿蛋白排泄显著降低(P<0.01),处死前大鼠血清白蛋白含量明显升高(P<0.01);肾脏病理分析示两商陆治疗组大鼠肾脏病理损害明显减轻,肾小球硬化指数显著减少(P<0.01);两治疗组Scr、BUN较模型组均有下降,其中商陆高剂量组与模型组差异显著(P<0.01)。结论商陆水煎剂能减少阿霉素肾小球硬化大鼠尿蛋白的排泄,减轻肾脏病理损害,保护肾功能。Objective To observe the protective effects of Pokeberry root decoction on glomerulosclerosis rats. Methods Forty - eight male SD rats were randomly divided into four groups : sham - operated group, model group, high - dose Phytolaccae - treated groups, and low- dose Phytolaccae - treated group. The glomerulosclerosis rats model was established via uninephrectomy and twice injections with adriamycin 5 mg · kg^-1 · d^ -1 ,3 mg· kg^-1 · d^-1 intravenously on the 'Tth and 28th day after uninephrectomy respectively. On the subsequent days the rats were respectively gastric - perfused treatment. The levels of 24 h urinary protein excretion and Scr, BUN and characteristic changes about renal tissue morphology damage were observed and the glomerulosclerosis index was calculated at the same time. Resuits At the end of 8th, 13th week after operation, The levels of 24 h urinary protein excretion were decreased in treated groups ,comparing with that in the model group(P 〈0.01 ) ; The levels of Scr in two Phytolaccae groups and BUN in high - dosage Phytolaccae group were lower than that in model group(P 〈0.01 ) ; The mean levels of BUN in low - dosage Phytolaccae group were significantly lower than that in model group though low - dosage Phytolaccae group was similar to model group( P 〉0.05 ); Renal tissue morphology damage in glomeruli, Glomerulosclerosis index in decoction - treated groups were weaker than that in model group ( P 〈 0.05 ) ; Conclusions Pokeberry root decoction can decrease24 h urinary protein excretion and improve renal tissue morphology damage , protect renal function.
