摘  要:目的:观察补肾复方胶囊含药血清对成骨细胞增殖、分化和矿化的影响。方法:从新生SD大鼠颅骨获取成骨细胞进行培养,取第三代大鼠成骨细胞,分别予以5%、10%、20%浓度的含药血清和10%对照血清培养,测定成骨细胞的增殖率、碱性磷酸酶活性、骨钙素含量、矿化结节数目。结果:与对照组比较,除3d、6d的低浓度组外,其它各组、各时间点均可增加成骨细胞增殖,差异有统计学意义(P<0.05或0.01);除低浓度组培养24 h外,各浓度给药组培养24 h、48 h、72 h后,碱性磷酸酶分泌均呈不同程度增加(P<0.05或0.01);各浓度均可增加骨钙素的分泌(P<0.05或0.01),尤以高剂量组为显著(P值均<0.01);中、高浓度可以显著促进矿化结节形成,差异有统计学意义(P<0.05)。结论:补肾复方胶囊含药血清在体外可以促进成骨细胞的增殖、分化和矿化,为其治疗骨质疏松提供实验依据。Objective: To explore the effects of pharmacological serum containing Bushenfufang Capsule on the proliferation, differ- entiation and mineralization of osteoblasts in vitro. Methods: The osteoblasts were taken from the cranial bones of newborn SD rats. After these osteoblasts had been cultivated, the third generation osteoblasts were taken and treated with different concentrations of pharmacological serum (5%, 10%, 20% ) and with control serum ( 10% ) respectively in order to detect cell proliferation assay, alkaline phosphatase (ALP) activity, osteocalcin content and mineralization. Results: When comparing with the control group, the proliferation of ostcoblasts could be significantly promoted in different groups at different time points except in the low concentration group on the 3rd day and 6th day time point ( P 〈 0. 05 or 0. 01 ) . Except for the low concentration group on the 24th hour time point, the secretion of ALP could be improved with various degrees (P 〈 0. 05 or 0.01 ) in different concentration groups at the time points of 24h, 48h, 72h. The secretion of osteocalcin could be improved in all the concentration groups ( P 〈 0. 05 or 0. 01 ), especially the high concentration group (P 〈 0. 01 ) ; mineralization could significantly be improved in the medium and high concentration groups ( P 〈 0. 05 ) . Conclusion: Pharmacological serum containing Bushcnfufang capsule could promote the proliferation, differentiation and mineralization of osteoblasts in vitro, which could be taken as the experimental evidence of treating osteoporosis.
