摘 要:目的:将犬骨髓间质干细胞(BMSCs)分离、扩增,观察生长情况,评价其作为组织工程种子细胞的可行性。方法:采用密度梯度离心及贴壁筛选法分离犬BMSCs,培养扩增,通过免疫细胞化学方法进行鉴定,Hoechst33342标记后传代,检测细胞荧光。结果:分离的BMSCs经免疫细胞化学方法观察到SH2-ir、Vimentin-ir、α2-SMA-ir细胞,标记后细胞生长旺盛,荧光无衰减。结论:BMSCs可以作为构建组织工程的种子细胞。Objective: To observe the growth status of canine bone marrow mesenchymal stem cells (BMSCs) and evaluate the feasibility as seeding cell. Methods: BMSCs were isolated and purified by Percoll density gradient centrifugation and by adhering to the culture plastic. After succeed culture and amplification, the morphology of cell was observed by immuocytochemistry staining. After la- beled by Hoechst33342, BMSCs were cultured. Results: SH2-ir, Vimentin-ir and α2-SMA-ir cells were observed in naturally differentia-t ed BMSCs. BMSCs labeled by Hoechst33342 showed good proliferation ability. Conclusion: The phenotypes and biological characteristics of canine BMSCs indicated that BMSCs was suitable to be used as seed-cells in constructing tissue-engineering.
