摘　　要：目的:建立微柱离心-HPLC法测定两性霉素B脂质体包封率。方法:采用葡聚糖凝胶(Sephadex G-50)微柱离心法分离脂质体与游离药物;用高效液相色谱法测定脂质体中两性霉素B的含量,计算包封率。结果:在所选色谱条件下,辅料不干扰测定,两性霉素B浓度在1.0～50.0mg·L-1范围内线性良好。微柱离心法能有效的将两性霉素B脂质体与游离药物分离,加样回收率为100.2%,RSD为0.97%。结论:该方法便捷,准确,重现性好,可用于测定两性霉素B脂质体的包封率。Objective：To establish a mini-column centrifugation HPLC method to determine the entrapment efficiency of amphotericin B liposome.Methods：A mini-column centrifugation was employed to separate the drug free from the liposome.The content of amphotericin B was quantified by HPLC.Results：Under the chromatogram condition the supplementary was applied with no influence.A calibrated linear curve of amphotericin B concentration was within 1.0-50.0 mg·L-1（r=0.9998）.The free amphotericin B was well separated from the liposome using mini-column centrifugation.The addition of blank liposome recovery rate of amphotericin B was 100.2% with RSD of 0.97%.Conclusion：The method was simple,accurate,reproducible,and applicable for determination of entrapment efficiency of amphotericin B liposome.
