摘  要:目的:建立原儿茶酸、酪醇、3,4-二羟基苯乙醇和3,4-二羟基苯甲酸的含量测定方法,对不同生产厂家炎立消制剂进行质量评价。方法:采用RP-HPLC法同时测定原儿茶酸和酪醇的含量:色谱柱为Dikma Technologies C18分析柱(200mm×4.6mm,5μm);流动相为乙腈-0.1%磷酸水溶液(6∶94),流速为1.0mL.min^-1;程序切换检测波长:0~18min时以258nm测定原儿茶酸的含量,18~30min时以278nm测定酪醇的含量。采用紫外-可见分光光度法,在500nm处测定吸收度,以原儿茶酸计测定3,4-二羟基苯乙醇和3,4-二羟基苯甲酸的含量。结果:HPLC法测定原儿茶酸的进样量在0.080~0.400μg范围内与峰面积呈良好的线性关系(r=0.9995),平均回收率为100.0%(RSD为2.1%,n=5);酪醇的进样量在0.630~3.150μg范围内与峰面积呈良好的线性关系(r=0.9995),平均回收率为99.3%(RSD为2.3%,n=5)。结论:用综合指数法评价炎立消制剂中多指标成分的含量,能够比较全面地控制其质量。Objective:To establish the method for determination of protocatechuic acid,tyrosol,3,4-dihydroxyphenethyl alcohol and 3,4-dihydroxybenzoic acid,and evaluate the quality of preparation of Yanlixiao from different manufacturers.Methods:Determination of protocatechuic acid and tyrosol by RP-HPLC,the chromatogra-phic column was Dikma Technologies C18 analytical column(200 mm×4.6 mm,5 μm),the mobile phase was acetonitrile-0.1%phosphoric acid solution(6∶94),the flow rate was 1.0 mL·min-1,the detection wavelength by programmable switching:0-18 min,at 258 nm for determination of protocatechuic acid,18-30 min,278 nm for determination of tyrosol.Determination of 3,4-dihydroxyphenethyl alcohol and 3,4-dihydroxybenz-oic acid that were calculated with protocatechuic acid by UV-Vis spectrophotometry at 500 nm.Results:A good linear range of protocatechuic acid in 0.080-0.400 μg by HPLC(r=0.9995),the average recovery was 100.0%(RSD was 2.1%,n=5);A good linear range of tyrosol was 0.63-3.15 μg by HPLC(r=0.9995),the average recovery was 99.3%(RSD was 2.3%,n=5).Conclusion:Assess multi-index ingredients in preparation of Yanlixiao with comprehensive index method can be used for the total quality control.
