摘  要:为筛选适于紫斑牡丹体胚发生的诱导培养基,以‘书生捧墨’花后90 d的种胚为外植体,采用L9(34)正交设计,研究了诱导培养基中基本培养基、蔗糖、BAP和NAA 4个因素及其组合对体胚发生的影响。结果表明:4个因素对体胚诱导率的影响均达到极显著水平,且以NAA对其影响最大,基本培养基和BAP次之,蔗糖的影响最小;4个因素的最佳组合为:改良MS+BAP 0.5 mg/L+NAA 0 mg/L+蔗糖100.0 g/L;经包含该组合的培养基培养70 d,体胚诱导率达63.3%,并获得了体胚再生植株。In order to screen optimal medium for inducing the somatic embryogenesis in Paeonia rockii hybrids,the effects of basal media,sucrose,BAP and NAA as well as their combinations on the inducement and development of somatic embryos were investigated from the embryos obtained at the 90(th) day after flowering based on the orthogonal design L_9(3~4).The results showed that all four factors had highly significant effects on the percentage of the explants with somatic embryos.The effect of NAA was the most significant,followed by those of basal media and BAP,and the effect of sucrose was the least significant.The optimum combination of the four factors was revised MS+BAP 0.5 mg/L+NAA 0 mg/L+Sucrose 100.0 g/L.Somatic embryos and regenerated plantlets were obtained in 63.3%of the explants cultured on the media with the optimum combination for 70 days.
