摘 要:目的探讨葡萄糖对胰岛β细胞系NIT-1上11β-羟类固醇脱氢酶1型(11β-HSD1)的表达及其功能的影响。方法用细胞免疫化学染色、RT-PCR和Western blot法检测NIT-1细胞11β-HSD1mRNA及蛋白表达。分别用含10、15、20和25 mmol/L葡萄糖的Ham s F12培养液培养NIT-1细胞72 h,检测11β-HSD1mRNA和蛋白表达水平,用葡萄糖刺激释放(GSIS)实验评价胰岛β细胞功能。结果(1)胰岛NIT-1细胞胞质里有棕黄色阳性染色颗粒,并有11β-HSD1mRNA和蛋白表达,(2)15、20和25 mmol/L葡萄糖组11β-HSD1表达增加(P<0.05,P<0.01),葡萄糖刺激胰岛素分泌减少(P<0.05,P<0.01)。结论11β-HSD1在胰岛NIT-1细胞上有表达,糖皮质激素可能参与胰岛β细胞胰岛素分泌功能。Objective To detect the glucose effect on the expression and function of 11 β-hydroxysteroid dehydrogenase type 1 ( 11 β-HSD1 ) in pancreatic beta cell line NIT-1. Methods The intraeellular localization of 11β- HSD1 was determined by immunoeytochemisty in NIT-1 cell. The expression of 11β-HSD1 mRNA and protein were analyzed by RT-PCR and Western blot. The NIT-1 cells were cultured for 72 hours in Ham's F12 medium with glucose concentration as 10 mmol/L, 15 mmol/L,20 mmol/L and 25 mmol/L. The function of beta cell was evaluated by glucose-stimulated insulin secretion (GSIS) test. Results ( 1 ) The distribution of 11β-HSD1 in cytoplasm was stained in brown. The expression of 11β-HSD1 mRNA and protein were identification in NIT-1 cell. (2) With increasing of glucose in medium, the expression of 11β-HSD1 increased and the secretion of insulin decreased in NIT-1 cell. Conclusion The pancreatic beta cell line NIT-1 expresses 11β-HSD1 gene. It is suggested that local glucocorticoid metabolism in pancreatic islet may regulate beta cell function.
