摘  要:目的:探讨中波紫外线(UVB)对正常小鼠表皮细胞凋亡及凋亡相关基因Bax和Bcl-2的调控。方法:将正常ICR小鼠随机分为对照组(假照射组)、不同剂量紫外线照射后2 h组。1500 J/m^2紫外线照射不同时间组。采用SP法染色检测凋亡相关基因Bax和Bcl-2的蛋白表达。结果:小鼠皮肤用不同剂量紫外线照射2 h后,Bax的表达随紫外线剂量增大而变大,Bcl-2表达减少,呈剂量依赖性。用1500 J/m^2紫外线照射小鼠后,Bax和Bcl-2的表达与紫外线照射后时间有关。结论:中波紫外线照射小鼠皮肤后,Bax表达上调,Bcl-2表达下调,促进小鼠紫外线损伤的表皮细胞凋亡。Objective: To investigate regulative effects of ultraviolet B (UVB) for normal apoptosis of epidermal cell in mice and apoptosis- related gene Bax and Bcl - 2. Methods: Normal ICR mice were divided into control group (sham irradiation group) and ultraviolet irradiation groups with dosage of 1000 J/m2, 1500 J/m2 and 2000 J/ m2 for 2 hours, and 1500 J/m2 for4, 8, 12, 24, 48, 72 hours. There were 10 mice in each group. Protein expression of apoptosis - related gene Bax and Bcl - 2 was detected by immunohistochemical method with streptavidin - pemxidase. Results: Bax expression increased and Bcl - 2 expression decreased in mouse skin after UVB irradiation with different dosages for 2 hours and both was in a dosage - dependant manner. Bax expression approached to peak value after UVB irradiation with 500 J/m2 dosage for 12 hours, whereas Bcl - 2 expression decreased. Bax expression began to decreased and Bcl - 2 expression increased after 24 hours, and returned to normal value after 72 hours. Bax expression and Bcl - 2 expression were related to time 'after UVB irradiation. Conclusion: Bax expression is increased and Bcl - 2 expression is decreased in mouse skin after UVB irradiation, which promotes apoptosis of epidermal cells injured by ultraviolet.
