摘  要:目的:探讨姜黄素固体分散体对酒精性肝损伤大鼠氧化应激的影响.方法:将50只SD大鼠随机分为5组:对照组、模型组、联苯双酯阳性药物组(100mg/kg)、姜黄素固体分散体低、高剂量组(100、200mg/kg),每组10只.除对照组给予等量生理盐水外,其余灌胃给予白酒与脂肪混合乳剂建立酒精性肝损伤模型,造模的同时给予联苯双酯,姜黄素固体分散体等药物干预.给药6wk后,测定大鼠血清中ALT,AST活性,测定肝组织中MDA含量,SOD活性,用以评价肝损伤程度及氧化应激反应程度.结果:与对照组比较,模型组大鼠肝质量/体质量值显著升高(3.74±0.38vs2.25±0.13,P<0.01);血清中ALT,AST水平显著升高(76.24±9.14vs30.13±4.99,98.85±10.50vs39.02±6.79,均P<0.01);肝组织中SOD活性显著降低(395.39±28.26vs258.18±22.18,P<0.01);MDA含量显著升高(27.00±1.08vs12.31±1.26,P<0.01).与模型组比较,各治疗组血清AST,ALT水平(AST:29.01±9.24,55.92±5.49,40.94±4.54;ALT:41.69±3.67,57.17±10.88,52.59±10.17)均显著降低(P<0.01).给予姜黄素固体分散体低、高剂量组肝组织中SOD活性显著升高(340.50±36.26,365.20±36.18,均P<0.01),而MDA含量明显下降(15.76±0.68,13.19±0.72,均P<0.01),而Bifendate组无显著性变化.结论:姜黄素固体分散体对酒精诱导大鼠肝损伤具有保护作用,并能降低肝损伤大鼠氧化应激反应.AIM: To prepare curcumin solid dispersion and to investigate the effects of curcumin solid dispersion on oxidative stress of alcoholic hepatic injury in rats.METHODS: Fifty Sprague-Dawley rats were randomly divided into 5 groups (n = 10): control group, model group, Bifendate group and two curcumin solid dispersion treatment (100, 200 mg/kg, respectively) groups. The SD rats were administrated with liquor and fat emulsion for 6 weeks to establish the alcoholic hepatic injury model, at the same time the rats were adminis- trated Bifendate and curcumin solid dispersion. The content of alanine aminotransferase (ALT), asparate aminotransferase (AST) in serum and the activity or content of superoxide dismutase (SOD) and malondialdehtyde (MDA) in liver were measured to evaluate hepatic injury and oxidative stress.RESULTS: Compared with control group, the value of liver weight/body weight was significantly increased (3.74 ± 0.38 vs 2.25 ±0.13; P 〈 0.01); the serum ALT and AST were significantly increased (76.24 ± 9.14 vs 30.13 ± 4.99; 98.85 ± 10.50 vs 39.02 ± 6.79; P 〈 0.01) but the activity of SOD was significantly decreased (395.39 ± 28.26 vs 258.18 ± 22.18, P 〈 0.01) and the content of MDA was significantly increased (27.00 ± 1.08 vs 12.31 ± 1.26, P 〈 0.01) in the model group. However, in comparison with those in model group, the serum AST and ALT were significantly decreased (Bifendate 29.01 ± 9.24, 41.69 ± 3.67; Low dose 55.92± 5.49, 57.17 ± 10.88; High dose 40.94 ± 4.54, 52.59± 10.17) in the two treatment groups. For low dose and high dose curcumin solid dispersion groups, activity of SOD was significantly increased (Low dose 340.50 ± 36.26, High dose 365.20± 36.18, P 〈 0.01) and content of MDA was significantly decreased (Low dose 15.76 ± 0.68, High dose 13.19 ± 0.72, P 〈 0.01) in liver, but there was no significant change in Bifendate group.
