新城疫病毒地方分离株HN基因的克隆及遗传变异分析

摘  要:对河北省保定地区两个NDV分离株用RT-PCR技术扩增其HN基因片断,产物经转化、克隆和测序,参照国内外已发表新城疫代表株绘制NDV系统发育进化树并分析其遗传关系,结果表明,两个分离株HN基因在遗传上同源性高达99.7%,两个分离株HN基因核苷酸序列与国内外报道的相关序列同源性为82.8%~93.2%,推导氨基酸序列同源性为90.0%~96.0%,由系统发育树可见,与现今流行于国内的Ⅶ型病毒株有密切联系。Two strains of Newcastle disease viruses isolated from Bao ding district in Hebei province were amplified by RT-PCR. The PCR products were purified, cloned and sequenced. According to the strains of the NDV reported home and abroad, the phylogenetic tree of the NDV and the analysis of genetic variation were obtaind. The result indicated that the homology of HN gene nucleotide sequences of 2 strains was 99.7% high. The HN gene sequences from two isolates shared a homology between 82. 8%-93.2% with that of related sequences reported home and abroad. The deduced amino acid sequences, shared a homology between 90.0%-96.0% with that of other strains. The phylogenetic tree showed that the two isolates and the demestic epidemic strains were closely related.
