摘  要:以西瓜纯种自交系伊选为材料,研究了化学诱变剂NaN3对西瓜直接再生试验体系的影响。研究发现,随着叠氮化钠浓度的升高,显著的延迟了种子萌发、胚根发生、子叶开展、不定芽分化、伸长以及生根的时间,降低了芽分化频率和每个外植体分化的不定芽数。1.5mmol/L的NaN3振荡处理1.5h为最佳的诱变处理,使外植体再生频率降低到40.0%,不定芽数/外植体降低到4.3,符合抗冷突变体半致死剂量的要求。Pure watermelon inbred line "Yixuan" was used in this experiment, the effects of NaN3 on the direct regeneration system of watermelon was studied. The results showed that with the increase of NaN3 concentration, the time of germination, radicle elongation, cotyledon extending, adventitious bud differentiation and elongation, and root regeneration was deferred distinctly; the percentage of adventitious bud differentiation and the ratio of adventitious bud differentiation per explant was lowered too. The best concentration for inducing chilling resistant mutant in "Yixuan" was 1.5 mmol/L, which decreased the frequency of explant regeneration to 40.0%, and the adventitious buds per explant to 4.3. It is accorded with the requirement of LDso for chilling resistant mutant.
