摘 要:应用游离小孢子培养技术对兼具有A,B,C染色体组的芸薹属异源六倍体CGMCCNo.2553(AABBCC,n=27)材料进行游离小孢子培养。结果表明,供试材料具有胚胎发生能力,并获得DH再生植株。220份驯化后的再生植株定植于日光温室中自交留种,其中12株获得自交种子。分别将DH植株与3份大白菜品种正、反交授粉杂交,得到4份杂交种F1代。经田间植物学鉴定,获得的杂交种为真正的杂种。目前,4份杂交种的染色体组成、倍性、育性等正在研究中。所获得的DH植株为今后构建遗传图谱,标记和定位重要的基因等后续研究奠定了基础。Crossing with Chinese cabbage ( Brassiea campestris L. ssp. Pekinensis ), Brassica allohexaploid CGMCC No. 2553 ( AABBCC, n=27 ), which owns A, B and C chromosomes simultaneously, was used for isolated microspore culture. The results showed that it could form embryoid and could get regenerated plants. 220 regenerated plants were grown at the greenhouse without heating apparatus. 12 DH plants got seeds by self-pollination. Through crossing DH lines with 3 Chinese cabbage pure lines, 4 F, hybrids were obtained. These hybrids were genuine hybrids by field checkup. They will be used for linkage map construction, locating and cloning the important genes in the future.
