黄连素和牛磺酸对胰岛素抵抗大鼠脂肪组织PAI-lmRNA表达的影响

摘　　要：目的：观察黄连素与牛磺酸对胰岛素抵抗（IR）大鼠的协同治疗作用。方法：SD雄性人鼠，按体重随机分为正常组和造模组，造漠组大鼠饲以特殊高脂饲料30d，造成胰岛素抵抗动物模型；以二甲双胍为对照，经黄连素和牛磺酸分别及联合治疗30d后，取血检测血糖、胰岛素、血脂等指标，计算（IR）敏感指数。运用RT—PCR技术测定大网膜脂肪组织PAI—1mRNA的表达。结果：采用特殊高脂饲料可以成功诱导胰岛素抵抗动物模型；黄连素和牛磺酸可分别提高胰岛素抵抗大鼠胰岛素敏感指数，降低TG，TC，LDL-C，升高HDL-C（P〈0．05），下调脂肪组织PAI—1mRNA的表达；黄连素和牛磺酸的协同作用优于单独作用。结论：黄连素与牛磺酸协同具有降糖、降脂、提高胰岛素敏感性作用，阻断PAI-1mRNA的过度表达的综合效应。Objective：To observe the effects of berberine and taurine on PAI-1mRNA expression in epiploon adipose tissue of high fat rats model of insulin resistance （IR）. Methods：SD rats were divided into normal and model groups at random by weight. Model groups were fed with high fat diet for 30 days firstly,screened the rats whose ISI decreased as model rats and treated with berberine and taurine respectively and combinedly for 30 dyas. After treatment,using RT-PCR to PAI-1mRNA from adipose tissue,the products were obtained,once its seqence was verified, semi quantitative RT-PCR employed to study its expressions in adipose tissues of rats with different groups. The fasting plasma glucose ,insulin,TG,TC and HDL-C were performed in each groups. ISI was calculated. Results：The IR model rats were induced by high fat diet. The berberine and taurine group and combine group could significantly increase insulin sensitivity （P〈0.05） ,decrease the weight （P〈0.05） ,and decrease the expression of PAI-1mRNA （P 〈0.05） in model rats. Meanwhile TG,TC,LDL-C decreased and HDL-C increased. The parameters mentioned above in combine group was better than other groups. Conclusion：Application of combining berberine and taurine is effective in increasing insulin sensitivity IR in rats induced by high-fat diet and decreasing the expression of PAI-1mRNA in adipose tissues. It had significant effect on changing the total effect of insulin resistance.
