摘 要:目的:建立反相高效液相色谱法测定苦胆草片中龙胆苦苷的含量。方法:色谱柱:Agilent XDB-C18柱(150mm×4.6mm,5μm);流动相:甲醇-水(30:70);流速:0.8ml.min-1;检测波长:270nm。结果:龙胆苦苷浓度在0.064~0.322mg.ml-1范围内呈良好的线性关系(r=0.9998),平均回收率为99.9%(n=5),精密度的RSD=0.58%(n=5)和重复性的RSD=0.43%(n=5)。结论:方法可靠,简单可行,为控制苦胆草片的内在质量提供了科学依据。Objective:To establish a method for the determination of gentiopicrin in Kudancao Tablets by RP-HPLC. Methods :The Agilent XDB C ls column ( 150 mm×4.6 mm, 5 μm) was used. The mobile phase was methanol-water(30:70).The flow rate was 0.8 ml·min^-1, and the detecting wavelength was at 270 nm.Results:Gentiopicrin had a good linearity in the range of 0.064-0.322 mg·ml^-1 (r=0.9998), and the average recovery of gentiopicrin was 99.9%(n=5).However, this method also had better precision(RSD=0.58% ,n=5) and good repro- ducibility (RSD=0.43%, n=5 ). Conclusion: This method is simple, reliable, sensitive and accurate.For its good specificity, this quantitative method provides a good scientific example for internal quality control of Kudancao Tablets.
