MMP-9和TIMP-1在育龄期DUB患者子宫内膜组织中的表达及意义

摘　　要：目的研究育龄期功能失调性子宫出血(DUB)患者子宫内膜局部基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制剂(TIMP-1)的表达变化。方法采用免疫组化S-P法检测35例育龄期DUB患者(病例组)及20例正常月经健康女性排卵后期(对照组)子宫内膜MMP-9、TIMP-1的表达情况。同时用TUNEL原位标记技术精确测定子宫内膜功能层的细胞凋亡指数。结果育龄期DUB女性以排卵型为主(30/35),其子宫内膜组织中MMP-9在出血部位的表达显著高于正常部位及对照组(P<0.05),TIMP-1在出血部位的表达显著低于正常部位及对照组(P<0.05),且出血部位细胞凋亡指数较正常部位及对照组显著增高(P<0.01),差异均有统计学意义。结论子宫内膜局部MMP-9与TIMP-1失衡与育龄期DUB的发生有关,细胞凋亡也参与了DUB的发生。Objective To observe the changes of expression of MMP-9 and TIMP-1 in endometrial region of women with dysfunctional uterine bleeding （DUB）. Methods The expression of MMP-9 , TIMP-1 in endometria was detected by immunohistochemistry in 35 patients with adult dysfunctional uterine bleeding and 20 healthy women（control group）. TUNEL technology was used to label in situ apoptotic cells to detect exactly apoptosis index in endometrial tissue. Results The expression of MMP-9 in local bleeding region was significantly higher than that in normal position of endometrium and that in control group. The expression of TIMP- 1 in local bleeding region was significantly lower than that in normal position of endometria and that in control group. Cellular apoptosis index in the bleeding region of endometrium was significantly higher than that in normal position of endometrium and that in control group. Conclusion The imbalance of MMP-9 and TIMP-1 in local endometrium region is related to the pathogenesis of adtdt dysfunction uterine bleeding. The cell apoptosis may be involved in the pathogenesis of DUB.
