摘 要:目的:对体外传代培养出现复制性老化的大鼠软骨细胞进行硫酸氨基葡萄糖干预实验,观察硫酸氨基葡萄糖对软骨细胞复制性老化的影响。方法:将第3代软骨细胞分为空白对照组、硫酸氨基葡萄糖不同浓度组,传代使细胞进入第4代,同时以第2代软骨细胞为青年对照组,进行组化检测老化相关β-半乳糖苷酶、流式细胞仪分析细胞周期和增殖指数、阿力新蓝染色检测胞外基质硫酸GAG(glycosaminoglycan)含量和结构、RT-PCR(reverese transcript-poly-merase chain reaction)检测Ⅱ型胶原及Aggrecan蛋白,并对硫酸氨基葡萄糖抗软骨细胞老化进行分子生物学研究。结果:硫酸氨基葡萄糖显著抑制老化相关β-半乳糖苷酶的表达(P<0.01),促进鼠软骨细胞增殖,减少G1期细胞含量,促进软骨细胞胞外基质GAG、Ⅱ型胶原、Aggrecan蛋白表达(P<0.01)。结论:硫酸氨基葡萄糖具有显著的抗软骨细胞复制性老化作用。Objective: To observe the effect of glucosamin sulphate (GS) on replicative senescence of rat chondrocyte subcultivated in vitro by means of GS interfering and controlled experiment. Methods : The 3^rd passage chondrocytes were divided into the blank group, different concentration GS groups passaged to 4^th generation, the control group of the 2nd passage. The chondrocytes of different groups were detected with the method of histochemistry for S - A - β - gal, for cell life cycle and proliferation index with flow eytometry, for the content and constructure of GAG of ECM with alcian blue test, and for type 11 collagen and Aggrecan with RT - PCR. A molecular biology study was also conducted on GS'preventive role against chondrocyte senescence. Results : GS can significantly inhibit chontrocyte's expression of S -A -β -gal, promote chontrocyte's proliferating( P 〈 0.01 ), reduce cell content in G1 phase, increase coment of GAG, and enchance the expression of type Ⅱ collagen and Aggrecan of ECM ( P 〈0.01 ). Conclusion: GS can significantly oppose rat chondrocyte senescence occurred in subcultivation.
